Aequorin bioluminescence assays on MicroBeta2 LumiJET for studying Ca2+-coupled GPCRs and ion channels

چکیده

Reconstitution of an active aequorin, using native coelenterazine or its derivative coelenterazine h, yields an enzyme having a fast luminescent response to increasing calcium concentrations, and a high level of signal intensity. The aequorin photoprotein undergoes a bioluminescent reaction in the presence of calcium ions, producing a flash of light peaking at 469 nm. This wavelength correlates well with the maximum quantum efficiency of the PMTs used in the MicroBeta2®. Directed Improvement of i-Photina Bioluminescence Properties, an Efficient Calcium-Regulated Photoprotein

Photoproteins are excellent reporter systems because they don’t have virtually background signal. Aequorin is the most well-known photoprotein. Three improved engineered photoproteins photina, i-photina and c-photina, were also recently developed and optimized for generation of Ca2+ mobilization assays precisely. The total light emission is greater than aequorin and their reacti...

Crystal structure of a Ca2+-discharged photoprotein: implications for mechanisms of the calcium trigger and bioluminescence.

Ca2+-regulated photoproteins are members of the EF-hand calcium-binding protein family. The addition of Ca2+ produces a blue bioluminescence by triggering a decarboxylation reaction of protein-bound hydroperoxycoelenterazine to form the product, coelenteramide, in an excited state. Based on the spatial structures of aequorin and several obelins, we have postulated mechanisms for the Ca2+ trigge...

Chimeric G proteins extend the range of insect cell-based functional assays for human G protein-coupled receptors.

We previously described a functional assay for G protein-coupled receptors (GPCRs) based on stably transformed insect cells and using the promiscuous G protein Galpha16. We now show that, compared with Galpha16, the use of chimeric Galphaq subunits with C-terminal modifications (qi5-HA, qo5-HA, or qz5-HA) significantly enhances the ability of insect cells to redirect Gi-coupled GPCRs into a Gq-...

Rapid ionic modifications during the aequorin-detected calcium transient in a skinned canine cardiac Purkinje cell

A microprocessor-controlled system of microinjections and microaspirations has been developed to change, within approximately 1 ms, the [free Ca2+] at the outer surface of the sarcoplasmic reticulum (SR) wrapped around individual myofibrils (0.3-0.4 micron radius) of a skinned canine cardiac Purkinje cell (2.5-4.5 micron overall radius) at different phases of a Ca2+ transient. Simultaneously mo...

Different NaCl-Induced Calcium Signatures in the Arabidopsis thaliana Ecotypes Col-0 and C24

A common feature of stress signalling pathways are alterations in the concentration of cytosolic free calcium ([Ca2+]cyt), which allow the specific and rapid transmission of stress signals through a plant after exposure to a stress, such as salinity. Here, we used an aequorin based bioluminescence assay to compare the NaCl-induced changes in [Ca2+]cyt of the Arabidopsis ecotypes Col-0 and C24. ...
